Reconstitution and concentration calculations

Semax in solution is refrigerated at 2 to 8 degrees and given a life of eight weeks to twelve by Australian Research Peptides' published condition. Semax opens on a methionine — MEHFPGP places the most oxidation-prone residue first, at an unblocked N-terminus — so light and dissolved air deserve as much attention as the calendar. Semax needs bacteriostatic water because the vial contains dry peptide and nothing wet. Semax is a synthetic peptide of seven amino acids, made by attaching a three-residue tail to a four-residue fragment of adrenocorticotropic hormone — the fragment chosen because it carries none of the parent hormone's hormonal activity. It was developed in Moscow and is studied in cognition and neuroprotection research, largely in Russian-language literature.

Published work on Semax centres on gene expression in brain tissue. Dmitrieva 2010 found Semax raised transcription of the genes for BDNF, nerve growth factor and their Trk receptors in rats. Those proteins keep neurons alive and maintain their connections. Dolotov 2006 gave rats a single intranasal dose of 50 micrograms per kilogram and saw hippocampal BDNF protein rise 1.4-fold. Sudarkina 2021 measured more active CREB, a gene switch, in damaged tissue.

Research on Semax is almost entirely in animals, and mostly about the brain after injury. Dmitrieva 2010 found that in a rat stroke model Semax activated transcription of Bdnf, Ngf and Nt-3 and their receptors. Sudarkina 2021 reported active JNK down more than 1.5-fold and Fos mRNA down 2.6-fold in ischaemic rat tissue. Eremin 2005 measured 5-HIAA, a serotonin breakdown product, up 25 percent in mouse striatum within two hours.

Australian Research Peptides supplies Semax as one freeze-dried solid in a sealed vial, not as a liquid nasal product. That solid is the chain MEHFPGP, at the milligram weight printed on the label. The powder runs white to off-white inside sealed, tamper-evident glass. There is no filler, no carrier, no second peptide and no liquid. Four of the seven amino acids reproduce part of adrenocorticotropic hormone, and the closing proline-glycine-proline is added in the lab.

Animal studies gave Semax into the nose or by injection into the abdomen. Dolotov 2006 used one intranasal dose of 50 micrograms per kilogram in rats. Sudarkina 2021 and Medvedeva 2014 both injected Wistar rats with 100 micrograms per kilogram, repeated over the first hours after an induced stroke. Eremin 2005 used 0.15 milligrams per kilogram by injection in C57BL mice. Every amount quoted for Semax comes from an animal study.

Semax survives in blood only briefly, though no clean half-life figure has been printed. Potaman's 1991 study in Biochemical and Biophysical Research Communications followed it through rat serum by chromatography, finding aminopeptidase stripping the first two residues away. The intermediates accumulated in comparable amounts, and those authors argued such fragments may carry part of the parent's activity, so the original chain disappearing is not the end of the account.

Semax is built from seven amino acids in one chain, which makes it a heptapeptide. In order they are Met-Glu-His-Phe-Pro-Gly-Pro, shortened to MEHFPGP. Met-Glu-His-Phe is a stretch of adrenocorticotropic hormone, positions four through seven. The last three are a synthetic addition the hormone itself does not have. The count of seven belongs to Semax itself, so both vial sizes contain the identical chain.

Semax has more human data behind it than most research peptides, because it has been a registered product in Russia for decades and its literature includes clinical studies in stroke and cognitive settings. That literature is largely Russian-language and has not been replicated in large Western trials, which is the main reason it remains unfamiliar elsewhere.

Semax's published studies sampled effects over hours to days rather than months, and the acute stroke work administered it over a defined short course. There is no long-run controlled trial establishing what happens over weeks of continuous use, so a timescale in months is not something the evidence supports either way.

Semax has decades of Russian clinical use behind it, and that literature describes courses with a defined endpoint rather than indefinite use, yet published observation after a course concludes is thin. No withdrawal syndrome is reported in it. Since that record has never been assessed outside Russia and large independent trials have not repeated it, it makes weak ground for confident statements about stopping.

Semax's registered form in Russia is an intranasal solution, and its clinical record describes it as well tolerated with local nasal irritation the main issue noted. No Western regulator has reviewed that record, and none of it describes the lyophilised research powder Australian Research Peptides supplies or any route other than nasal.

Storage, condition and testing

Semax comes in two quantities and they look markedly different. Ten milligrams leaves a faint dusting easily read as an empty container; thirty milligrams is three times as much solid and forms a properly visible cake. Neither holds a bulking agent, so what sits inside the glass is peptide alone. Comparing the two strengths explains most confusion about the smaller one.

Store the sealed Semax vial frozen, dry and out of the light. GenScript flags cysteine, methionine and tryptophan as prone to oxidation, and Semax opens with a methionine, which is why light and air matter. Bachem keeps freeze-dried peptides below minus 15 degrees Celsius in a closed container, and GenScript's own figure is minus 20, away from bright light. Warm a frozen vial fully before you break the seal, so moisture does not condense inside. Peptide in solution is far less stable than the dry powder.

Most cloudy Semax reports come down to bubbles worked in during mixing, and they lift out within minutes of the vial being set down. The thirty-milligram strength dissolves more slowly than the ten, having three times as much material to take up, so patience beats extra agitation. Once warmed and rested a proper preparation is clear; lasting milkiness or floating specks are not normal.

Semax is analysed two ways before supply — HPLC first, then mass spectrometry. HPLC separates the sample and reports what fraction runs as the main peak. Mass spectrometry reads the molecular mass and compares it against what the sequence predicts. For Semax that second check matters, because it separates a seven-amino-acid chain from the shorter ACTH(4-7) fragment it opens with. No matching certificate is currently published for this product and strength.

Delivery, payment and access questions

Semax is sold in Australia as a laboratory research chemical, and Australian Research Peptides supplies it on that basis only. ARP makes no medical claim for Semax anywhere. Australia sorts medicines and poisons into schedules under the Poisons Standard, administered by the TGA. Anything to do with use in a person sits outside what ARP supplies Semax for.

Semax has never been approved in the United States. It is approved somewhere, however: Russia's health ministry registered it as a medicine in 1994, in an intranasal form. A registration granted in one country is not a worldwide licence, and no Western agency has assessed the Russian-language clinical record standing behind it. Freeze-dried research powder is not that registered product in any case.

Semax has no listing of its own in any prohibited category. Whether the residual S0 heading reaches it turns on one point: Russia's health ministry registered Semax as a medicine in 1994, and S0 excludes anything a government regulator has approved somewhere. Whether an anti-doping panel accepts a Russian registration is untested, so Australian athletes should ask Sport Integrity Australia.
