## What Is Ipamorelin?

Ipamorelin is a fully synthetic peptide of five amino acids, Aib-His-D-2-Nal-D-Phe-Lys-NH2, classed as an agonist of the growth hormone secretagogue receptor. It was developed at Novo Nordisk under the code NNC 26-0161 and first described in 1998.

Three of its five building blocks are not found in ordinary proteins, two are mirror-image D-forms, and the chain finishes in an amide rather than an acid. That sequence occurs in no human protein: ipamorelin was designed from scratch rather than cut from a natural chain.

## Which Receptor Does Ipamorelin Act On?

Ipamorelin acts on the growth hormone secretagogue receptor, also known as the ghrelin receptor: it binds the site that the hormone ghrelin normally occupies and switches the receptor on. A secretagogue is anything that prompts a cell to release a substance, and here the cells are the growth-hormone-producing cells of the pituitary.

The 1998 paper by Raun and colleagues that introduced the compound is titled “Ipamorelin, the first selective growth hormone secretagogue”. Its authors placed the activity at a GHRP-like receptor, and worked in three systems: primary rat pituitary cells, anaesthetised rats and conscious pigs.

## What Does the Research Literature Study Ipamorelin For?

Published ipamorelin research follows two strands: how the peptide drives growth hormone release in pituitary cells and animals, and how it affects movement through the gut in rat models.

- **Growth hormone release.** The original characterisation measured the growth hormone response in isolated rat pituitary cells, in rats and in swine. It reported on the receptor and the response, not on any outcome in a person.
- **Gastrointestinal motility.** A 2009 study by Venkova and colleagues asked whether ipamorelin would speed gastrointestinal transit in fasted male rats after abdominal surgery and handling of the intestine, a model of postoperative ileus.

Ipamorelin has no marketing approval in any country. A clinical programme was run and later discontinued, so the published record is the laboratory and animal work above.

## Is Ipamorelin the Same as the CJC-1295 and Ipamorelin Blend?

Ipamorelin on its own is not the same product as the CJC-1295 (no DAC) + Ipamorelin blend: the single vial holds ipamorelin only, while the blend dries two different peptides together in one vial.

The partner peptide in the blend, CJC-1295 without DAC, works through a different receptor — the GHRH receptor — which is why the two are paired rather than treated as alternatives. ## How Is Ipamorelin Tested?

Ipamorelin is tested by HPLC, which measures what share of the powder runs as a single peak, and by mass spectrometry, which checks that the molecule weighs what a five-residue chain with an amide end should weigh.
